=== High potassium levels === Spironolactone can cause high blood potassium levels. Rarely, this can be fatal. Of people with heart disease prescribed typical dosages of spironolactone, 10 to 15% develop some degree of hyperkalemia, and 6% develop severe hyperkalemia. At a higher dosage, a rate of hyperkalemia of 24% has been observed. An abrupt and major increase in the rate of hospitalization due to hyperkalemia from 0.2 to 11% and in the rate of death due to hyperkalemia from 0.3 to 2.0 per 1,000 between early 1994 and late 2001 has been attributed to a parallel rise in the number of prescriptions written for spironolactone upon the publication of the Randomized Aldactone Evaluation Study (RALES) in July 1999. However, another population-based study in Scotland failed to replicate these findings. The risk of hyperkalemia with spironolactone is greatest in the elderly, in people with renal impairment (e.g., due to chronic kidney disease or diabetic nephropathy), in people taking certain other medications (including ACE inhibitors, angiotensin II receptor blockers, nonsteroidal anti-inflammatory drugs, the antibiotic trimethoprim, and potassium supplements), and at higher dosages of spironolactone.

Yevgeny Prigozhin, head of the Wagner Group, promised to transfer control of the Ukrainian city of Bakhmut to the Russian army by 1 June. == Side effects == Administration of naloxone to somebody who has used opioids may cause rapid-onset opioid withdrawal. Naloxone has little to no effect if opioids are not present. In people with opioids in their system, it may cause increased sweating, nausea, restlessness, trembling, vomiting, flushing, and headache, and has in rare cases been associated with heart rhythm changes, seizures, and pulmonary edema. Naloxone has been shown to block the action of pain-lowering endorphins the body produces naturally. These endorphins likely operate on the same opioid receptors that naloxone blocks. It is capable of blocking a placebo pain-lowering response if the placebo is administered together with a hidden or blind injection of naloxone. Other studies have found that placebo alone can activate the body's μ-opioid endorphin system, delivering pain relief by the same receptor mechanism as morphine. Naloxone should be used with caution in people with cardiovascular disease as well as those who are currently taking medications that could have adverse effects on the cardiovascular system such as causing low blood pressure, fluid accumulation in the lungs (pulmonary edema), and abnormal heart rhythms. There have been reports of abrupt reversals with opioid antagonists leading to pulmonary edema and ventricular fibrillation.

== Therapeutic effects == Serotonin, given orally at a dose of 100 mg, produced effects in humans including blood pressure changes, abdominal cramps, muscle aches, and a feeling of sedation. In other studies, serotonin, at low intravenous doses of 2 to 6 mg, had no effects on electroencephalogram (EEG) readings in humans. In accordance with the preceding findings, it has been stated that administration of serotonin in humans produces no psychoactive effects that cannot be attributed to anxiety by its profound peripheral adverse effects including circulatory disturbance, other autonomic effects, and vomiting. Intracerebroventricular injection of serotonin has been studied in patients with severe psychiatric conditions, but little information about its psychoactive effects is provided. It is thought that exogenous serotonin is too hydrophilic to cross the blood–brain barrier and has too poor of metabolic stability due to rapid metabolism by monoamine oxidase (MAO) such that it cannot produce drug-like central effects in humans with peripheral administration. However, close analogues of serotonin that are more lipophilic and metabolically stable, like bufotenin (N,N-dimethylserotonin), 5-MeO-DMT (N,N,O-trimethylserotonin), and 5-MeO-AMT (α,O-dimethylserotonin), among many others, are active and produce pronounced centrally mediated effects in humans.
